Separation of Human and Various Animal Lipoproteins by Vertical Agarose Gel Electrophoresis

Lipoprotein electrophoresis is in common use in the clinical laboratory to aid the classification of the different forms of dyslipoproteinemias. In lipoprotein electrophoresis, the various support media such as paper, agarose, starch, polyacrylamide and cellulose acetate are utilized. Today, the majority of the electrophoresis is performed on ready made gels purchased from commercial companies, and for the most part, the most popular lipoprotein electrophoresis support medium at this time is agarose gel. The agarose system separates the pre-β lipoprotein fraction from the β lipoprotein with better resolution than other electrophoretic systems. Agarose gel electrophoresis has been described historically horizantal. In this study, firstly, horizantal electrophoresis system was adapted to vertical system, and secondly serum lipoprotein patterns [HDL (a), LDL (β) and VLDL (pre-β) ] were determined in the normal cattle, sheep, goat, cat, dog, horse and chicken using the vertical agarose gel electrophoresis and compared with that in humans. In the vertical agarose gel electrophoresis of the human lipoproteins, the three classes of lipoproteins (β, pre-β and a lipoproteins) were detected. Agarose gel electrophoresis of the lipoproteins showed two major serum lipoprotein fractions in the animals studied, except the chicken, as β (+ pre-β) and a lipoproteins. In the animals, both pre-β and β lipoproteins migrated on agarose to approximately the same position. But in a dog, β, pre-β and a bands on electrophoresis were seen. In the agarose gel electrophoresis of the chicken serum, pre-βlipoprotein migrated only a short distance from the origin and a lipoprotein at the trailing edge of albumin. In the chickens (Broiler ) β lipoprotein could not be determined. It was concluded that the vertical agarose gel electrophoresis might be used as a reference method in the evaluation of dyslipoproteinemias of human and various domestic animals.
