Effect of L-Arginine Addition on Short Term Storability of Ram Semen

This study was conducted to investigate the effect of L-arginine addition on short term storability of ram semen. The semen collected from 6 Akkaraman rams by using artificial vagina once a weeks during 5 weeks, was pooled. The semen samples were diluted with Tris-egg yolk extender to include 400 million motile sperms per mL and were then divided into 6 equal groups. While group 1 was kept as control, 0.1 mM, 0.5 mM, 1 mM, 5 mM, 10 mM L-arginine were added to the other groups. The groups were kept at +4 ⁰C for short term storage. Motility, membrane integrity, arginase activity were analyzed at 0, 12th, 24th h and then at 24 h intervals. When the samples stored in short term at +4 ⁰C were evaluated, it was observed that the addition of 10mM L-arginine to semen samples decreased the motility after the 12nd h and membrane integrity after the 48th h (P<0.05), but the addition of 10 mM L-arginine decreased the arginase activity at the 24th, 72th, 96th (P<0.01). In conclusion, 10 mM dose of L-arginine added to the diluent damaged to the quality of the semen during short-trem storage.
