Determination of Respiratory Coronavirus Viral Antigens by Immunohistochemistry and Polymerase Chain Reaction (PCR) in Cattle Lungs with Pneumonia in Elazığ Region

In this study, it was aimed to determine the presence and prevalence of Bovine Corona Virus (BCoV) by histopathological, immunohistochemical and Reverse-transcription-polymerase chain reaction (PCR) methods in the lung samples with pneumonia taken from cattle in the Elazig region. Tissue samples taken from 363 cattle lungs showing signs of pneumonia macroscopically were examined histopathologically. In the lungs with microscopic interstitial pneumonia; Thickening of the interalveolar septum, mononuclear cell infiltration, desquamation of the bronchial epithelium, bronchioalveolar lymphoid tissue (BALT) hyperplasia, bronchiolitis obliterans and syncytial cell formations were recorded. Immunohistochemical and PCR analyzes were performed on the lungs that were evaluated as interstitial pneumonia. Immunohistochemically, BCoV viral antigens were detected in 24 lungs. Specific staining due to viral antigens was detected more intensely in the bronchi and bronchiolar epithelium, peribronchioalveolar lymphoid tissue, and to a lesser extent in mononuclear cells in the interstitial region, macrophage and syncytial cells in the interalveolar space, and cellular exudate in the bronchiolar lumen. No positivity was detected in the amplicons of clinical samples in PCR analyzes performed only on interstitial pneumonia samples. DNA fragments of the expected size (406 bp in length) were seen in the amplicons of the BCoV positive controls in each reaction.
