Introduction: In this study, the association between Reeline and migraine disease was investigated through analyzing reelin gene polymorphism in patients with migraine with aura. Migraine is the most common vascular headache in which genetic and environmental components are acting and which leads to loss of labor force. Reelin gene localized on chromosome 7 synthesizes an extracellular matrix protein which plays a role in central nervous system development. Any mutations or altered expressions in reelin gene are known to precipitate neuropsychiatric diseases.

Methods: Ninety-one patients diagnosed with migraine and 85 healthy controls were included in the study. DNA was obtained in blood samples and rs7341475, rs362691, rs12705169 polymorphisms were studied with RT-PCR using TaqMan probes.

Results: rs7341475 polymorphism and A allele detected in migraine patients were statistically significant when compared with control group. There was a significant correlation between nausea and rs12705169 (P<0.05). It was found a significant association between sudden and slow onset of headache and rs362691 (P<0.05).A difference was not detected in genotype distribution of three SNP polymorphisms with regard to gender and habits (like smoking) (P>0.05).

Conclusion: Our study showed that there is a significant association between migraine and Reelin gene rs7341475 polymorphism and A allele.
