The Role of Kisspeptin-10 on Oxidant and Antioxidant Activity in the Kidney in the Mice Model of Transient Cerebral Ischemia

Objective: While the individual effects of ischemia after carotid artery occlusion and kisspeptin on the oxidative stress are known, there is no research in the literature on the effect of kisspeptin on oxidative stress in peripheral tissues after carotid artery ischemia (CAI). In this study, it was aimed to investigate the effect of kisspeptin given before ischemia on oxidative alterations in kidneys after ischemia.

Materials and Methods: Adult C57/BL6 mice were divided into 4 groups as 1-Sham, 2-Kisspeptin, 3-Ischemia and 4-Ischemia+Kisspeptin (n=5). Groups were administered only saline physiologic (Sham and Ischemia groups) and kisspeptin (Kisspeptin and Ischemia+Kisspeptin groups) intraperitoneally 40 minutes before ischemia and the operation (sham). Subsequently, the Ischemia groups were exposed to CAI for 15 minutes and reperfused for two hours. After the 175 min experimental period, animals were decapitated, and kidneys were removed, total oxidant and antioxidant status (TOS and TAS, respectively) were determined.

Results: Carotid artery ischemia did not cause any changes in TOS and TAS parameters. The kisspeptin administration to ischemic group caused an insignificant increase in TAS levels (P=0.41), while no effect was observed on TOS levels. There was no effect on TAS levels, while TOS levels were insignificantly reduced in the Ischemia+Kisspeptin group compared to sham operation (P=0.18). TAS was significantly higher in the Ischemia+Kisspeptin group compared to Kisspeptin group (P=0.032).

Conclusion: There was no change in TAS and TOS levels after CAI. However, the increase in total antioxidant status upon kisspeptin administration without causing any change in the total oxidative status suggests that kisspeptin increases antioxidant activity in the kidneys.
