Telomerase Enzyme Activity Observed In Urıne Samples of Bladder Cancer Patıents

Telomerase is a reverse transcriptase formed by its own RNA and proteins,and is also responsible from telomere length control. Telomere is a sort of special DNA series with heterochromatine structure occuring at both ends of eucaryot cell chromosomes. Telomerase activity is essential for the immortality of tumor cells, and for the manipulate of carcinogenic process. Therefore this activation is consideration as an indicator for the diagnosis of cancer. Also telomerase activation exhibits an increase in proportion to the development of cancer, has brought attention to the idea that it may be a marker for monitoring the course of the tumour.

This study was planned to determine telomerase in the cells of the urines of bladder cancer patients.For this goal, we used 7 male without diagnosed bladder cancer in control group and 13 male diagnosed with bladder cancer and without chemotheraphy in patient group who were referred to the urology clinics of Eskişehir Osmangazi University Medical Faculty. The mean age all of the experiment groups were over the 40 years old. The telomerase of cell extracts were determined by using TRAP method. We used the TeloTAGGG TelomerasePCR –ELİSAPLUS (Roche) kit. PCR products, electrophoresis process was applied, and then bands were made noticeable by silver painting. Statistical analysis, it was found that telomerase was higher in urinary bladder cancer patients than the control group. This findings indicate that telomerase could aid in the diagnosis of bladder cancer, and also could help in following the response of the patients to treatment.
