Investigation of MDM2 Gene Expression Changes in Peripheral Blood Lymphocytes After In-Vitro Gamma Radiation Exposure

Objective: The murine double minute 2 (MDM2) negatively controls the p53 tumor suppressor gene and destroys the p53 with E3 ubiquitin ligase activation. In this study, effect of DNA damage caused by different in vitro gamma radiation doses in peripheral human lymphocytes on MDM2 gene expression was investigated.

Material and Methods: Gene expression changes were evaluated with quantitative real time polymerase chain reaction (QRT PCR) using B2M as a reference gene. For this reason, whole blood samples of ten young, healthy and non-smoking donors were collected. The first tube of the blood samples which were divided in five tube for each individual, was used as control and other four tubes were irradiated using a constant gamma irradiation source (60Co) giving the doses of 0.5 Gy, 1 Gy and 3 Gy, which were calculated according to dose rate of 0,0327 kGy/h.

Results: Results were expressed in terms of the threshold cycle value (CT) in REST 2009 (Relative Expression Software Tool V. 2.0.13) program and the difference between the CT values of the target gene and the reference gene was calculated.

Conclusions: QRT PCR results showed that MDM2 gene was up-regulated after radiation exposure. The results of the present study suggest that MDM2 gene can be used to determine the effects of exposure to ionized radiation.
