Protective Effect of Humanin on the Lungs by Myocard Infarct Induced Rats: The Role of TRPM2

Objective: In this study, the potential role of transient receptor potential (TRP) melastatin-like subfamily member 2 (TRPM2) in its protective effect on the lungs in MI by administering humanin (HN) to rats before myocardial infarction (MI) was investigated.

Materials and Method: Rats were divided into 7 groups (n=6) (control, 2 groups of HN, 2 groups of MI, 2 groups of MI+HN). To induce MI, 200mg/kg isoproterenol (ISO) was administered subcutaneously to rats. Rats were given 2 mg/kg HN as a single agent intraperitoneally (ip) before MI. TRPM2 level in lung tissue was examined immunohistochemically.

Results: In the study, TRPM2 expression was observed in the smooth muscle structures and interalveolar areas of vessels and bronchioles in the lung tissue. It was observed that TRPM2 levels increased especially towards the 7th day after MI, and decreased more significantly towards the 7th day after HN administration before MI. In the groups where only HN was administered, no change was observed in terms of TRPM2 levels.

Conclusion: In this study, it was concluded that HN has a protective effect on the lungs in MI and this effect may be mediated by TRPM2
