Development of Skin During Prenatal and Postnatal Stages In Rats

The aim of the present study was to determine the development of paraffin rat skin on prenatal days 13, 17, 19, 21 and on neonatal day 3 several histochemical staining methods. The thick sections of the rat skin were stained with hematoxylene-eosin, Grimelius, Mallory-Azan and periodic acid-Schiff (PAS) reaction o88n prenatal days 13, 17, 19, 21 and neonatal day 3. Skin samples obtained in prenatal days 17, 21 and neonatal 3 were fixed in 3 % gluteraldehyde, postfixed in 1% osmium tetroxide and embedded in Araldite CY 212. Semi-thin sections were stained with toluidin blue and evaluated under the light microscope.

On day 13 of life, epidermis had 2 layers, periderm and basal layer. While periderm was lined with single layer squamous epithelium, basal layer was lined with single layer cuboidal epithelium. The layer between dermis and epidermis, consisting of a loosely packed mesenchymal cells, was a straight line. On the day 17 of fetal life, epidermis had three distinct cell layers, basal, dikensi and granular cell layers respectively. Callagen and elastic fibers abundant hair follicles and capillaries were seen in the dermis. Prepapillar structures developing in the epidermis-dermis border were also seen on the 17.

On the day 19 of intrauterine life while epidermis layer seemed similar to the mature skin, collagen fibers and capillaries increased in the dermal layer along with papillar processes starting to be distinguished in the epidermis-dermis border. On prenatal day 21, epidermis developed a fiber like keratin layer. Dermis had internal and external root sheats of hair follicle which was similar to the mature skin. On neonatal day 3, epidermis seemed to have its characteristic stratified squamous keratinized epithelium whereas dermis had increased blood capillaries, connective tissue fibers, hair follicles and abundant adipocytes.
