Cloning of ORF2b Gene of TT Virus

Objective: Transfusion-transmitted virus (TTV) was first identified in patients with non-A to -G hepatitis following blood transfusion in 1997. The objective of this study is cloning of ORF2b gene fragments of TTV.

Materials and Methods: In this study, the ORF2b genes from TTV DNA positive samples were amplified by polymerase chain reaction (PCR) and cloned into pGEM®-T plasmid vector.

Results: The recombinant plasmid vectors including ORF2b gene were detected by PCR-screening test and restriction enzyme analysis.

Conclusion: The producing of the gene clones containing ORF2b gene of TTV will contribute the enrichment of the viral gene library of our country. Moreover, the target proteins produced from the recombinant clones in future studies will be used in terms of the development of ELISA kits.
